RAPID COMMUNICATION Differential Redistribution of Platelet Glycoproteins Ib and IIb-IIIa After Plasmin Stimulation

چکیده
The subcellullar localization of the platelet membrane receptors glycoproteins (GP) Ib and Ilb/lla has been studied within resting platelets by a combination of biochemical and cytochemical techniques. While both GPlb and GPllb/llla are localized within the plasma membrane and surface-connected canalicular system (SCCS) membranes, only GPIIb/ llla is present within the internal face of a-granular membranes. Previous studies demonstrated that plasmin can induce platelet stimulation and also decrease ristocetininduced platelet aggregation; it was suggested that this was because of GPlb degradation by plasmin. In this study, the respective localizations of both GPlb and GPllb/llla were visualized during in vitro plasmin stimulation of platelets. Generally, plasmin induced shape change, pseudopod formation, organelle centralization either with or without a-granule release depending on the conditions of stimulation. Plasmin treatment of platelets at 37°C resulted in the disappearance of GPlb from the cell surface and its subsequent redistribu-

Differential redistribution of platelet glycoproteins Ib and IIb-IIIa after plasmin stimulation.
The subcellular localization of the platelet membrane receptors glycoproteins (GP) Ib and IIb/IIIa [corrected] has been studied within resting platelets by a combination of biochemical and cytochemical techniques. While both GPIb and GPIIb/IIIa are localized within the plasma membrane and surface-connected canalicular system (SCCS) membranes, only GPIIb/IIIa is present within the internal face ...
متن کاملEffects of leukocyte-derived cathepsin G on platelet membrane glycoprotein Ib-IX and IIb-IIIa complexes: a comparison with thrombin.
Cathepsin G is a serine, chymotrypsin-like protease released by activated polymorphonuclear leukocytes (PMN) that may act as a platelet agonist. The effect of this enzyme on platelet surface glycoproteins (Gp) Ib and IIb-IIIa was evaluated by means of a cytofluorimetric assay, using fluorescein isothiocyanate-labeled monoclonal antibodies (MoAbs) directed at the alpha chain of Gp Ib (SZ2), at G...
متن کاملThrombin-induced platelet membrane glycoprotein IIb and IIIa complex formation. An electron microscope study
The topographic relationships of platelet membrane glycoprotein IIb and glycoprotein IIIa have been studied in stimulated and unstimulated human platelets using immunoelectron microscopy. An indirect approach with ferritin-conjugated goat anti-rabbit gamma-globulin was used to localize the rabbit antibody to glycoprotein IIIa. The second ultrastructural label was keyhole limpet hemocyanin conju...
متن کاملStructural changes in platelet glycoprotein IIb/IIIa by plasmin: determinants and functional consequences.
Plasmin exposure modulates platelet aggregation responses, but a direct effect of plasmin on the platelet fibrinogen receptor, glycoprotein IIb/IIIa (GPIIb/IIIa), has never been conclusively shown in a plasma milieu. To examine this issue, we incubated platelets in platelet-rich plasma with plasmin and measured the effect of this treatment on platelet aggregation, fibrinogen binding, and the st...
متن کاملThrombin decreases von Willebrand factor binding to platelet glycoprotein Ib.
Thrombin is a physiological agonist that promotes platelet aggregation and secretion. In this study we observed that thrombin can also inhibit a function of platelets related to primary hemostasis. Platelet stimulation by thrombin decreased the binding of von Willebrand factor (vWF) to glycoprotein (GP) Ib and decreased ristocetin-induced agglutination, in vitro reactions that correlate with in...
